Product Specific Information

Description:
The RPA-T8 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as a heterodimer (CD8α/β) or homodimer (CD8α/α). CD8a is present on thymocytes, mature T cells, and NK cells. It binds to MHC class I and associates with protein tyrosine kinase p56lck, playing a key role in T-cell development and activation.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis using normal human peripheral blood cells. Recommended usage: 5 µL (0.125 µg) per test (100 µL final volume). Suitable for 10⁵–10⁸ cells/test.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples may be stored in IC Fixation Buffer (Product #00-822-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days in the dark at 4°C with minimal impact on brightness and FRET efficiency.

Target Information

CD8 (Cluster of Differentiation 8):
CD8 is a type I transmembrane glycoprotein of the immunoglobulin receptor family involved in signal transduction and T-cell differentiation. It is mainly expressed on T cells as a CD8α/β heterodimer acting as a co-receptor for MHC class I molecules. CD8α is essential for MHC-I binding and is also found on NK cells, monocytes, and dendritic cells as CD8α homodimers.
Binding of MHC-I/peptide complexes induces Lck recruitment, leading to lymphokine production and CTL activation. Activated CTLs are crucial for pathogen and tumor cell clearance. Differentiation into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
