Dynabeads Streptavidin for Target Enrichment beads are 1-μm monosized, monodispersed, superparamagnetic beads covalently coupled to streptavidin and intended for enrichment of target sequences for library preparations for next-generation sequencing (NGS) workflows.

Consistency and reproducibility
The monolayer of streptavidin leaves most of the biotin-binding sites available for the binding of free biotin and biotinylated ligands. The specific and defined surface allows for efficient capture, separation, and downstream handling. In addition, the low size distribution (<5%) of the monosized beads and the streptavidin monolayer help ensure negligible leakage, batch consistency, and reproducibility of results.

High binding capacity
The size of the molecule and the biotinylation procedure will affect the binding capacity. The capacity also depends on steric availability and charge interaction between bead and molecule and between molecules. With two or three biotin binding sites available for each streptavidin molecule on the surface of the bead after immobilization, one mg of Dynabeads Streptavidin for Target Enrichment typically binds:
• 950–1,500 pmol free biotin
• ∼20 μg biotinylated ds-DNA
• ∼400 pmol biotinylated ss-oligonucleotides

Applications
Thermo Fisher streptavidin-coupled Dynabeads magnetic beads have been used and cited for a wide variety of applications. Although the beads can be used to isolate any biotinylated targets, these hydrophobic beads are particularly well suited for the enrichment of target sequences for library preparations for NGS workflows. Dynabeads Streptavidin for Target Enrichment is a high-quality product that enables robust and reproducible NGS analysis results on any NGS platform.

Dynabeads Streptavidin for Target Enrichment efficiently captures DNA sequences of interest through specific hybridization via immobilization of target-specific biotinylated probes to the streptavidin on the bead surface. The subsequent washing steps utilize magnetic separation to obtain high purity. The starting sample can be nucleic acid extracted from chromatin immunoprecipitated samples (ChIP) or isolated pure nucleic acid.
