Target Information
Beta-catenin은 세포 간 접착 구조(adherens junction, AJ)의 구성 요소로 처음 확인된 단백질입니다. AJs는 세포 성장과 세포 간 접착을 조절하여 상피 세포층의 생성과 유지에 필수적입니다. Beta-catenin은 E-cadherin의 세포질 도메인과 상호작용하여 E-cadherin을 alpha-catenin에 연결하고, 이를 통해 cortical actin cytoskeleton에 부착시킵니다. 또한 adenomatous polyposis coli 단백질 및 미세소관을 포함한 세포골격 복합체와 결합하며, tyrosine kinase, phosphatase, Wnt/Wingless 등의 신호전달 경로와도 상호작용합니다. 이러한 상호작용은 형태형성(morphogenesis)을 조절할 수 있습니다.
Beta-catenin은 모낭 세포, 기저 및 주변 기질 세포, 외부 및 내부 뿌리초 세포 등 다양한 세포 유형에서 발현됩니다. 병리학적으로는 pilomatrixoma, medulloblastoma, colorectal cancer, ovarian cancer 등 여러 종양 발생과 관련이 있습니다. 핵 내에서 Beta-catenin은 Lef/Tcf 전사인자군을 공동활성화하여 cyclin D, c-myc 등의 유전자 발현을 촉진하며, 이는 세포 증식과 종양 발생에 중요한 역할을 합니다.
