Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells, comprising approximately 10% of surface proteins in lymphocytes.
It is absent in non-hematopoietic cell lines and tissues. CD45 glycoprotein plays a crucial role in lymphocyte development and antigen signaling, regulating Src-family kinases.
Multiple isoforms exist due to alternative splicing, differing in extracellular domains but sharing transmembrane and cytoplasmic domains.
CD45RA is one isoform with restricted expression among lymphoid subtypes. CD45 isoforms vary in membrane domain translocation and expression depending on cell type and physiological state.
It is an essential regulator of T- and B-cell receptor signaling and modulates cytokine receptor signaling through JAK kinase suppression.
Additionally, CD45 contributes to cell survival, integrin-mediated signaling, DNA fragmentation during apoptosis, and regulation of various immunological functions.
