Description:
The 26F8 monoclonal antibody reacts with human CD317 (BST2, tetherin). CD317 is a 30–36 kDa type II transmembrane protein expressed on B cells and bone marrow stromal cells. Although CD317 mRNA is detectable in activated T cells, protein expression in primary T cells and macrophages is undetectable. CD317 is associated with pre-B cell growth and plasma B cell differentiation. It also prevents HIV-1 virion release from infected cells, an activity inhibited by the HIV-1 Vpu protein. CD317 serves as the ligand for the ILT7 receptor.

Applications Tested:
Tested by flow cytometric analysis on MCF7 cells. Recommended ≤0.125 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Antibody titration recommended for optimal performance.

Use in a sterile environment. Filtered post-manufacturing (0.2 µm).

Target Information

BST2 (CD317, HM1.24 antigen, DAMP-2, tetherin):
An integral membrane protein involved in B-cell development and growth. BST2 expression on fibroblast lines supports stromal cell-dependent pre-B-cell line growth. Expressed in B-cell lineages, T cells, monocytes, NK cells, dendritic cells, and myeloma cells. Functions as an inhibitor of retrovirus release, antagonized by HIV-1 Vpu. BST2 depletion abolishes Vpu requirement for virus release, suggesting potential therapeutic relevance for HIV/AIDS. BST2 dysfunction is also linked to stomatitis.
