Product Specific Information

The OKT8 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as CD8αβ heterodimers or CD8αα homodimers. CD8a is found on thymocytes, mature T cells, and NK cells. It binds MHC class I and associates with p56lck to mediate T-cell development and activation.

This antibody has been validated for flow cytometric analysis of normal human peripheral blood cells.
Use at 5 µL (0.125 µg) per test (100 µL final volume). Cell numbers typically range from 10⁵–10⁸ cells/test.

Light Sensitivity: Protect vial and stained samples from light.
Fixation: Samples may be stored up to 3 days at 4°C in IC Fixation Buffer or 1-step Fix/Lyse Solution with minimal effect on brightness.
Excitation: 488–561 nm; Emission: 775 nm; Compatible with Blue, Green, Yellow-Green lasers.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family. It functions as a co-receptor with TCR for MHC class I molecules and is essential for T cell activation. CD8 is expressed on cytotoxic T cells, NK cells, monocytes, and dendritic cells. Upon MHC-I ligation, CD8 recruits Lck kinase, triggering cytokine production and cytotoxic T lymphocyte activation, crucial for pathogen and tumor clearance. Differentiation of naive CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
