Background

The spike (S) glycoprotein of coronaviruses is essential for viral binding to host cells during infection initiation. The SARS-CoV spike (S) glycoprotein mediates membrane fusion required for virus entry and cell fusion, serving as a major immunogen and a target for entry inhibitors.
2019-nCoV infects human respiratory epithelial cells via interaction with the human ACE2 receptor. The spike protein is a large type I transmembrane protein composed of two subunits, S1 and S2. S1 contains the receptor-binding domain (RBD) responsible for receptor recognition, while S2 contains elements required for membrane fusion.
The S protein plays key roles in inducing neutralizing antibodies, T-cell responses, and protective immunity.
