Target Information

Sclerostin is a glycoprotein predominantly secreted by osteocytes and belongs to the Cerberus/DAN family of BMP antagonists. It acts as an endogenous regulator of the canonical Wnt signaling pathway and inhibits bone homeostasis.
Although mainly expressed by osteocytes, sclerostin is also found in bone, bone marrow, cartilage, kidney, and liver, and can be produced by hypertrophic chondrocytes and cementocytes.
Sclerostin regulates the Wnt/β-catenin signaling pathway by forming inhibitory complexes with LRP5 and LRP6, essential co-receptors for Wnt ligands. It also interacts with LRP4 to further inhibit Wnt signaling and promotes osteoclast activity by increasing RANKL expression in osteocytes.
Mutations leading to reduced sclerostin levels cause bone dysplasia disorders such as sclerosteosis and van Buchem disease, characterized by progressive bone overgrowth and hypermineralization.
