Target Information

The CD3 subunit complex is crucial in transducing antigen-recognition signals into the cytoplasm of T cells and regulating the cell surface expression of the TCR complex. T cell activation through the TCR involves the cytoplasmic tails of CD3 subunits (CD3 gamma, CD3 delta, CD3 epsilon, and CD3 zeta). These subunits are structurally related members of the immunoglobulin superfamily encoded by closely linked genes on human chromosome 11.

CD3 components possess long cytoplasmic tails that interact with signal transduction molecules via a double tyrosine-based motif. CD3 may play roles in TCR-induced growth arrest, cell survival, and proliferation. The CD3 antigen is expressed on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum, but not on B or NK cells. Reduced T lymphocyte percentages may be observed in certain autoimmune diseases. Genes encoding CD3 epsilon, gamma, and delta are located on chromosome 11, and CD3 gene defects are associated with CD3 immunodeficiency.
