Target Information
Oxygen radicals damage chromosomal DNA, leading to cell death and mutations. Among the various types of oxidative DNA damage, 8-oxoguanine (8-oxoG) is a key lesion that can cause G:C to T:A transversion mutations.
8-oxoG and its nucleotide form 8-oxo-dGTP are produced during normal metabolism and oxidative stress. MTH1 (MutT homolog 1) hydrolyzes 8-oxo-dGTP to prevent its misincorporation into DNA, maintaining genomic integrity.
MTH1 also recognizes 8-oxo-rGTP, preventing its incorporation into RNA, which could otherwise result in defective protein synthesis.
