Target Information
ACAP2 is a transport carrier belonging to the Centaurin family with three ANK repeats, an Arf-GAP domain, a BAR domain, and a PH domain.
It functions as a GTPase-activating protein (GAP) for ADP ribosylation factor 6 (ARF6) and exhibits strong constitutive ARF6 GAP activity.
The activity is stimulated by phosphatidylinositol 4,5-bisphosphate (PIP2) and phosphatidic acid, with minimal activity toward ARF1.
ACAP2 regulates the return of ARF to the inactive GDP-bound state and, together with ACAP1, is targeted by phosphatidylinositol (3,4,5)-triphosphate, inhibiting PDGF-induced dorsal membrane ruffle formation in NIH3T3 fibroblasts when overexpressed.
It is phosphorylated upon DNA damage, likely by ATM or ATR, and shows widespread expression, highest in lung tissue.
