Target Information
ALAS-1은 glycine과 succinyl-CoA의 축합 반응을 촉매하여 δ-aminolevulinate, CO₂, CoA를 생성하는 핵 인코딩 미토콘드리아 효소입니다. 헴 생합성 경로의 첫 번째이자 속도 제한 단계의 조절 효소로, heme을 필요로 하는 cytochrome 및 hemoprotein에 공급합니다. 간에서 ALAS1은 heme에 의해 음성 피드백 조절을 받으며, cAMP 및 호흡 uncoupling에 의해 전사가 촉진되고 phorbol ester와 insulin에 의해 억제됩니다. ALAS1은 heme 생합성과 생체 리듬 간의 상호 조절에 관여하는 것으로 제안되며, 이는 생체 시계 장애 치료의 잠재적 표적이 될 수 있습니다. 급성 간부전 시 ALAS1 발현이 감소하여 heme 대사 및 간 기능에 영향을 미칩니다.
