Product Specific Information
A 1:1,000 dilution of PA5-77771 was sufficient for detection of KDEL-containing proteins in 20 µg of HeLa cell lysate by ECL immunoblot analysis using goat anti-mouse IgG as the secondary antibody.
Detects KDEL proteins, GRP94, Grp78, PDI, and calreticulin. It may also detect ERp57 and ERp29.
Target Information
The sequence Lys-Asp-Glu-Leu (KDEL), or closely related variants, is located at the carboxy-terminus of soluble endoplasmic reticulum (ER) resident proteins and some membrane proteins.
78 and 94 kDa glucose-regulated proteins (GRP78 and GRP94) and protein disulfide isomerase (PDI) share the C-terminal KDEL sequence.
This sequence is critical for ER retention and can reduce secretion of proteins from the ER.
Retention is mediated by a KDEL receptor.
