Target Information
Estrogen Receptors (ER) belong to the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor acts as a ligand-activated transcription factor that, upon binding to estrogen, undergoes conformational changes allowing dimerization and DNA binding to estrogen response elements (ERE). ER regulates gene transcription positively or negatively via coactivator or corepressor recruitment.
There are two forms of estrogen receptors, alpha (ESR1) and beta (ESR2), encoded by distinct genes. Alternative RNA splicing results in multiple isoforms of estrogen receptor-alpha (66 kDa, 53 kDa, 47 kDa, 35 kDa). These receptors are expressed in various tissues and are crucial for sexual development, reproduction, and bone maintenance. Dysregulation of estrogen receptors is implicated in diseases such as breast cancer, endometrial cancer, and osteoporosis.
