Target Information
TMP21 is a ubiquitously expressed protein involved in vesicular targeting and protein transport. It is also a component of the presenilin complex and modulates the γ-secretase, but not the ε-secretase, cleavage activity of the amyloid precursor protein. The presenilin complex includes APH1, nicastrin, PEN2, and presenilin-1, which together cleave amyloid precursor protein at γ- and ε-sites, potentially leading to accumulation of Abeta associated with Alzheimer’s disease. Co-immunoprecipitation experiments suggest TMP21 regulates this complex. Suppression of TMP21 by siRNA increases γ-secretase activity and Abeta production, demonstrating its modulatory role.
