Product Specific Information
- Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 min.
Target Information
Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein belonging to the immunoglobulin family of receptors. It plays a crucial role in signal transduction, T cell differentiation, and activation.
CD8 is predominantly expressed on T cells as a disulfide-linked heterodimer of CD8α and CD8β, functioning as a co-receptor with the T cell receptor (TCR) for MHC-I molecules.
CD8α is essential for MHC-I binding and is also found on subsets of T cells, NK cells, monocytes, and dendritic cells as homodimers.
Upon ligation with MHC-I/peptide complexes presented by antigen-presenting cells (APCs), CD8 recruits Lck, leading to lymphokine production, motility, and cytotoxic T lymphocyte (CTL) activation.
Activated CTLs are vital for pathogen and tumor cell clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
