A recommended positive control tissue for this product is skeletal muscle, though other tissues may also be used.
This antibody is intended for laboratory professional use in detecting Myoglobin in formalin-fixed, paraffin-embedded tissues using manual qualitative immunohistochemistry (IHC). It is used after the primary diagnosis of tumors by conventional histopathology.
Myoglobin Function and Diagnostic Use:
- Myoglobin is a cytosolic oxygen-binding protein responsible for oxygen storage and diffusion in myocytes.
- Expression is highest in skeletal and cardiac muscle.
- Acts as an O₂-storage protein, releasing oxygen during hypoxia or anoxia.
- Useful in identifying rhabdomyosarcoma and tumors with skeletal muscle differentiation.
- Reportedly expressed in epithelial cancer cells under altered metabolic conditions.
Staining and Controls:
- Use with formalin-fixed and paraffin-embedded sections.
- Pretreatment with heat-induced epitope retrieval or enzymatic retrieval recommended.
- IHC staining involves sequential application of primary antibody, secondary antibody, enzyme complex, and chromogenic substrate.
- A positive tissue control must be run with every staining procedure.
- Positive control tissues should contain both positive and negative staining components.
- External positive controls should be properly fixed and embedded specimens.
- Weakly positive tissues are recommended to detect subtle changes in antibody sensitivity.
- Internal or external negative controls may be used as per institutional guidelines.

This gene encodes a member of the globin superfamily, expressed in skeletal and cardiac muscle. The protein is a hemoprotein that contributes to intracellular oxygen storage and transcellular oxygen diffusion.
