Target Information
The 6x His tag is a synthetic oligopeptide consisting of six consecutive histidine residues (HHHHHH).
It is commonly expressed at N- or C-terminal regions of recombinant proteins to enable purification by immobilized metal affinity chromatography (IMAC).
Epitope tagging allows detection of recombinant proteins even when specific antibodies are unavailable.
His-tagged proteins are purified via metal chelate chromatography due to the strong affinity between histidine clusters and metal ions such as Ni²⁺.
His tag-specific antibodies are useful for detection and co-immunoprecipitation of tagged proteins.
