Target Information

CXCR4 (also known as Fusin, LESTR, HUMSTR) is a G-protein-coupled chemokine receptor with seven transmembrane domains. It functions as a co-receptor for X4 HIV-1 entry into CD4⁺ cells and has also been identified as a co-receptor for HIV-2 binding via gp120, facilitating Env-mediated viral fusion.
CXCR4 serves as the receptor for the chemokine CXCL12/SDF-1, mediating intracellular calcium signaling and MAPK1/MAPK3 activation. It also acts as a receptor for extracellular ubiquitin, influencing calcium and cAMP signaling.
CXCR4 plays essential roles in hematopoiesis, cardiac septum formation, and vascularization of the gastrointestinal tract. In the central nervous system, it contributes to hippocampal neuron survival and cerebellar development. Additionally, CXCR4 binds bacterial LPS and mediates LPS-induced inflammatory responses, including TNF secretion by monocytes.
Antibodies targeting CXCR4 can block HIV-1 and HIV-2 infection of human target cells. The N-terminal domain and the second extracellular loop of CXCR4 serve as HIV binding sites.
