Target Information

CD11a (LFA-1 alpha, ITGAL) together with CD18 constitute leukocyte function-associated antigen 1 (LFA-1), the alphaLbeta2 integrin.
CD11a is implicated in activation of the LFA-1 complex and plays a central role in leukocyte intercellular adhesion through interactions with ICAM-1, -2, and -3.
It also functions in lymphocyte costimulatory signaling and is expressed on the plasma membrane of leukocytes in a low-affinity conformation.
Upon stimulation, CD11a converts to a high-affinity conformation supporting tight binding to ICAMs.
CD11a is involved in immune cell interaction, tissue-specific settlement, and regulation of cell differentiation, proliferation, and T-cell effector functions.
Blocking CD11a function by antibodies or small molecules is a therapeutic approach for inflammatory diseases.
Efalizumab (Raptiva) is a humanized anti-CD11a antibody used to inhibit T-cell migration, while simvastatin binding to CD11a allosteric site modulates immune activity.
Two transcript variants encoding different isoforms of CD11a have been identified.
Diseases associated with CD11a dysfunction include benign cephalic histiocytosis and leukocyte adhesion deficiency.
