Product Specific Information

This eyedio monoclonal antibody reacts with human indoleamine-2,3-dioxygenase (IDO, INDO, IDO1), an intracellular enzyme that catalyzes the degradation of tryptophan to kynurenines. IDO is expressed in a wide variety of tissues and cells, including macrophages, plasmacytoid dendritic cells, and several cell lines. IDO can be induced by IFN-γ or other inflammatory stimuli. Expression of IDO in antigen-presenting cells and tumors mediates immune suppression via tryptophan depletion or creation of metabolites that induce T cell apoptosis and regulatory T cells.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and validated for intracellular staining using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00) and BestProtocols® Protocol A.
Use 5 µL (0.06 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm and is excited with the blue laser (488 nm). Recommended filter: 710/50 (alternative 695/40). Ensure instrument compatibility.

Light Sensitivity:
Protect from light to avoid photo-induced oxidation.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness.

Excitation / Emission / Laser:
488 nm / 710 nm / Blue Laser

Filtration:
0.2 µm post-manufacturing filtered.


Target Information

IDO1 is an intracellular heme-containing enzyme that catalyzes the oxidative cleavage of the indole ring of molecules such as tryptophan, serotonin, and melatonin, leading to the production of kynurenines. IDO1 is expressed in various human tissues, macrophages, and dendritic cells. Induced by interferons, IDO1 participates in antimicrobial defense and immunoregulation during infection, pregnancy, autoimmunity, transplantation, and tumorigenesis.
