Target Information
CD20 is a non-glycosylated surface phosphoprotein with a molecular weight of 33–37 kDa, depending on phosphorylation. It is expressed on mature and most malignant B cells, as well as in a subset of T lymphocytes and follicular dendritic cells. CD20 expression correlates with surface IgM and regulates calcium conductance, cell cycle progression, and B-cell proliferation.
CD20 is associated with lipid rafts, with the strength of this association influenced by extracellular signals and conformational changes. Upon receptor ligation, CD20 and BCR colocalize transiently before BCR endocytosis, while CD20 remains on the cell surface. CD20 is a key target for antibody-mediated B-cell depletion therapies since it is highly expressed on B-cell malignancies and remains membrane-bound after treatment.
Diseases associated with CD20 dysfunction include Ms4a1-related common variable immune deficiency.
