Target Information
Glycogen synthase kinase-3 (GSK3)는 원래 glycogen synthase를 조절하는 단백질 키나아제로 확인되었으며, 단백질 합성, 세포 증식, 분화, 미세소관 조립/분해, 세포자멸사 등 다양한 세포 기능 조절에 관여합니다.
GSK3는 고유한 기질 특이성을 가지며, 인산화는 기질의 C-말단 4번째 잔기에 phosphoserine 또는 phosphotyrosine이 존재할 때만 일어납니다.
GSK3에는 α와 β 두 가지 isoform이 존재하며, 인산화에 의해 엄격하게 조절됩니다. Protein kinase B (PKB)에 의한 Ser9 (GSK3α의 Ser21) 인산화는 GSK3β의 불활성화를 유도하며, 이는 성장인자에 의한 억제의 주요 기전입니다.
활성화된 GSK3β는 p53, c-Myc, c-Jun, HSF-1, cyclin D1 등 다양한 단백질을 인산화하며, tau 단백질의 비정상적 인산화에도 관여하여 알츠하이머병의 진행과 관련이 있습니다.
