Background
Functions in nuclear protein import as a nuclear transport receptor. Serves as receptor for nuclear localization signals (NLS) in cargo substrates. Mediates docking of the importin/substrate complex to the nuclear pore complex (NPC) through nucleoporin binding, followed by translocation via an energy-dependent Ran mechanism.
At the nucleoplasmic side, Ran binding causes dissociation of the importin/substrate complex, and importin is re-exported to the cytoplasm. This process ensures directional nuclear import regulated by Ran-GTP/GDP distribution.
IPO9 mediates the nuclear import of H2B histone, RPS7, and RPL18A, preventing their cytoplasmic aggregation. It may also import H2A, H3, H4 histones, RPL4, and RPL6. Belongs to the importin beta family, containing one importin N-terminal domain. Binds with high affinity to RPS7 and RPL18A, and interacts with PPP2R1A and PPP2R1B.
