Target Information

Ephrin A5 is a cell surface GPI-bound ligand for Eph receptors, which are receptor tyrosine kinases crucial for migration, repulsion, and adhesion during neuronal, vascular, and epithelial development. It binds Eph receptors on adjacent cells, leading to contact-dependent bidirectional signaling.
Forward signaling occurs downstream of the receptor, while reverse signaling occurs downstream of the ephrin ligand. Ephrin A5 induces compartmentalized signaling within caveolae-like membrane microdomains requiring Fyn tyrosine kinase activity.
It activates the EPHA3 receptor to regulate cell-cell adhesion and cytoskeletal organization. In conjunction with EPHA2, it may regulate lens fiber cell shape and interactions, contributing to lens transparency. Ephrin A5 also stimulates axon fasciculation and mediates communication between pancreatic islet cells to regulate glucose-stimulated insulin secretion.
It is a cognate ligand for EPHA7, playing a role in brain development through modulation of cell-cell adhesion and repulsion.
