Target Information

Members of the Rho family of Ras-related GTPases, such as ARHE, regulate the organization of the actin cytoskeleton in response to extracellular growth factors. Like Ras, Rho family members cycle between inactive GDP-bound and active GTP-bound forms.
Three major regulators of Rho activity have been identified:

1. RhoGDIs – interact with GDP-bound Rho proteins to maintain resting state
2. GEFs – promote GDP/GTP exchange leading to activation
3. GAPs – stimulate GTP hydrolysis to return to inactive form
