Target Information
CD146 (MCAM) is an integral membrane glycoprotein belonging to the immunoglobulin superfamily. It is expressed at cell-cell junctions in all endothelial cells and in various normal and malignant cell types. CD146 functions as a calcium-independent cell adhesion molecule and may also participate in signal transduction via recruitment of Fyn kinase, leading to tyrosine phosphorylation of intracellular proteins involved in actin cytoskeleton assembly. It plays a key role in endothelial cell cohesion and vascular integrity, and its expression is associated with melanoma cell interaction with vascular components, enhancing tumor spread. Additionally, CD146 may act as an adhesion molecule in neural crest cells during embryonic development and as a surface receptor that triggers tyrosine phosphorylation of FYN and PTK2, causing transient intracellular calcium increase.
