Target Information
PACS-1 (CRA_a) is a 963 amino acid regulatory protein involved in protein trafficking and belongs to the PACS family. It controls the subcellular localization of trans-Golgi network (TGN) membrane proteins containing acidic cluster sorting motifs. PACS-1 is also implicated in HIV-1 Nef-mediated removal of MHC-I from the cell surface to the TGN, helping HIV-1 evade immune surveillance. Furthermore, PACS-1 regulates endosome-to-Golgi trafficking of furin and mannose-6-phosphate receptor by linking their acidic-cluster-containing cytoplasmic domains with the AP-1 adapter-protein complex of endosomal clathrin-coated pits. It is typically localized in the Golgi apparatus, trans-Golgi network, and paranuclear region, and is ubiquitously expressed in most tissues.
