Target Information
Ubiquitin is a 76 amino acid highly conserved eukaryotic polypeptide that selectively marks cellular proteins for proteolytic degradation by the 26S proteasome. The process of target selection, covalent attachment, and transport to the proteasome regulates the concentrations of key cellular proteins by limiting their lifespans.
Polyubiquitination involves serial enzymatic steps including activation by E1, transfer to E2, and conjugation to substrates via E2 or E3 ligase. Deubiquitination enzymes reverse this process.
Posttranslational tagging by ubiquitin participates in diverse cellular processes such as cell cycle regulation, differentiation, apoptosis, DNA repair, transcription regulation, transport, stress response, immunity, and neural functions.
