Target Information
CD20 is a non-glycosylated surface phosphoprotein with a molecular weight range of 33–37 kDa depending on phosphorylation. It is expressed on mature and most malignant B cells, a subset of T lymphocytes, and follicular dendritic cells. CD20 expression coincides with surface IgM expression and regulates calcium conductance, cell cycle progression, and B-cell proliferation. It associates with lipid rafts, with the interaction modulated by extracellular signaling or BCR aggregation. CD20 remains on the cell surface after BCR endocytosis and serves as a key target for antibody-mediated B-cell depletion therapies, as it is highly expressed on B-cell malignancies and is not internalized upon antibody binding. CD20 dysfunction is linked to Ms4a1-related common variable immune deficiency.
