Target Information

Members of the Id family of basic helix-loop-helix (bHLH) proteins include ID1, ID2, ID3, and ID4. They are ubiquitously expressed and form heterodimers with class A and B HLH proteins. Because ID proteins lack the basic DNA-binding region, these heterodimers cannot bind DNA, acting as negative regulators of bHLH transcription factors.
ID1 inhibits DNA binding of E12 and Myo D, thereby suppressing muscle-specific gene expression. During muscle differentiation, ID protein levels decrease, allowing E12/E47 to interact with Myo D and myogenin to activate myogenic differentiation. Expression of ID proteins is growth factor–dependent, and reduction of ID mRNA delays cell cycle reentry after growth factor stimulation.
