Product Specific Information

CD230 (prion protein; PrP^C) is a 35 kD glycoprotein attached to the cell surface via a GPI-anchor. Although its function remains unclear, prion protein is central to research involving transmissible spongiform encephalopathies (TSE), including Creutzfeld-Jakob disease in humans. CD230 undergoes a conformational change from PrP^C to PrP^Sc, which is essential for prion disease infectivity.

CD230 is mainly expressed on lymphocytes and monocytes, weakly on granulocytes and erythrocytes, and intracellularly in platelets—where it translocates to the surface upon activation.

Applications Reported:
The 4D5 antibody is suitable for flow cytometry and immunoprecipitation. It also recognizes a ~35 kD protein under non-reducing conditions in immunoblotting.

Applications Tested:
Validated by flow cytometric analysis of normal human peripheral blood cells. Recommended ≤0.5 µg/test (100 µL final volume). Optimal performance requires titration based on cell number (10^5–10^8 cells/test).

Target Information

Human prion protein (PrP, PRNP) is a GPI-anchored cell surface glycoprotein associated with lipid rafts and signaling functions. It plays roles in apoptosis, epithelial cell proliferation, and junctional protein distribution. Conversion from PrP^C (alpha-helical) to PrP^Sc (beta-sheet-rich, proteinase K-resistant) triggers prion-related neurodegenerative diseases such as Creutzfeld-Jakob disease (CJD).
