• Immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
• For formalin-fixed tissues, heat sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, then cool at room temperature for 20 min before staining.

Target Information

CD2 (LFA-2) is a monomeric surface antigen (MW 45–58 kDa) expressed on human T-lymphocytes and some natural killer cells, but not on B lymphocytes. It is one of the earliest T-cell markers, present on over 95% of thymocytes.

• CD2 interacts with CD58 to stabilize adhesion between T cells and antigen-presenting or target cells.
• Low affinity in solution is compensated by strong two-dimensional cell-cell adhesion.
• T cell activation increases CD2 expression and mobility, enhancing CD2–CD58 interactions.
• CD2 deficiency is linked to reduced intestinal inflammation and certain immune disorders.
• Diseases associated with CD2 dysfunction include penile squamous cell carcinoma and thymus-related immune deficiency.
