Product Specific Information

This P84 monoclonal antibody reacts with mouse CD172a (signal regulatory protein alpha, SIRPα).
CD172a is a cell surface glycoprotein with three Ig-like extracellular domains and two cytoplasmic ITIM motifs, which recruit SHP-1 and SHP-2 phosphatases.
It is expressed on monocytes, macrophages, and dendritic cells, but not on T or B lymphocytes. Neurons and CNS tissues also express CD172a.
CD47 serves as the extracellular ligand for CD172a, and this interaction influences dendritic cell-mediated T cell activation, neutrophil migration, and phagocytosis.
This monoclonal antibody has reported neutralizing activity.

Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometric analysis of mouse bone marrow cells, ≤0.5 µg/test (100 µL final volume, 10⁵–10⁸ cells/test).
Antibody titration is recommended for optimal performance.

Target Information

SIRP alpha (CD172a) is a receptor-type transmembrane glycoprotein expressed on myeloid lineage cells (granulocytes, dendritic cells, macrophages, mast cells, hematopoietic stem cells).
It functions as a substrate for tyrosine kinases such as EGFR, PDGFR, src, and insulin receptor, negatively regulating receptor tyrosine kinase signaling pathways.
Binding of SIRPα to CD47 leads to ITIM phosphorylation and SHP-1/2 recruitment, modulating immune responses including inhibition of phagocytosis and T cell/DC activation.
SIRPα also regulates cellular responses to serum, growth factors, insulin, oncogenes, and adhesion, playing a role in physiological and pathological processes.
Cancer cells often overexpress CD47, activating SIRPα to evade macrophage-mediated destruction.
