Product Specific Information

The PC61.5 antibody reacts with mouse CD25, the 55 kDa interleukin-2 receptor alpha chain (IL-2Rα). CD25 is expressed by early progenitors of T and B lineage and activated mature T and B lymphocytes. It binds IL-2 with low affinity alone but forms a high-affinity receptor when associated with CD122 (IL-2Rβ) and CD132 (common γ chain).

Binding of IL-2 to both receptor classes is inhibited by the PC61.5 antibody. CD25 plays a key role in lymphocyte differentiation and activation/proliferation.

• Applications Tested: Mouse splenocytes, ≤0.125 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration for best performance.
• Light Sensitivity: Tandem dye sensitive to photo-induced oxidation; protect from light.
• Fixation: Samples can be stored in IC Fixation Buffer (Product #00-822-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark.

Target Information

CD25 (IL2RA) encodes the interleukin-2 receptor alpha chain, part of a heterotrimeric receptor complex with IL2RB and IL2RG.

• High-affinity receptor: IL2RA + IL2RB + IL2RG
• Low-affinity receptor: IL2RA homodimer
• Medium-affinity receptor: IL2RB homodimer

IL2 is primarily produced by mature T cells and regulates proliferation, differentiation, and apoptosis of lymphocytes. It stimulates B cell growth, enhances NK cell activity, and suppresses granulocyte macrophage colony formation. Disruption of IL2RA in mice leads to ulcerative colitis-like disease, suggesting its role in immune response regulation. Mutations are associated with interleukin-2 receptor alpha deficiency.
