Product Specific Information

This AD2 monoclonal antibody reacts with human CD73, a 5’-ectonucleotidase that converts 5’-adenosine monophosphate (AMP) to adenosine. CD73 is expressed on endothelial cells, B cells, and T cells including CD4⁺Foxp3⁺ regulatory T cells. Adenosine production by these cells inhibits CD4 T cell effector functions such as proliferation and cytokine secretion.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells at 5 µL (0.4 µg) per test.
A test is defined as staining a cell sample in a final volume of 100 µL (10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

CD73 (Ecto-5'-nucleotidase) catalyzes the conversion of purine 5'-mononucleotides to nucleosides, preferentially AMP. It forms a dimer of two identical 70 kDa subunits bound to the plasma membrane via a glycosyl phosphatidylinositol linkage. CD73 serves as a lymphocyte differentiation marker; its deficiency is associated with immunodeficiency diseases. The CD73 gene is localized to chromosome 6q14–q21. Mutations can cause joint and arterial calcification and intestinal tuberculosis. Two transcript variants encoding different isoforms have been identified.
