Product Specific Information

Description: HI100 monoclonal antibody reacts with human CD45RA, a 220 kDa molecule expressed by subpopulations of CD4+ and CD8+ peripheral T lymphocytes and B cells. CD45RA+ T cells are mainly naive/virgin, allowing use as a phenotypic marker to discriminate T cell subsets.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use at 5 µL (0.03 µg)/test. A test is defined as the amount of antibody staining a cell sample in 100 µL final volume. Cell number may range from 10⁵ to 10⁸ cells/test.

Fluorophore Characteristics:
PE-eFluor® 610 is excited by 488–561 nm laser lines and emits at 607 nm. Recommended filter: 610/20 band pass (PE-Texas Red® equivalent). Ensure instrument compatibility.

Light Sensitivity: Tandem dye sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal brightness loss.

Target Information

CD45RA is an isoform of the CD45 complex with restricted expression among lymphoid cell subtypes. CD45 (leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells, comprising ~10% of lymphocyte surface proteins. It regulates Src-family kinases, influences lymphocyte development, and modulates antigen signaling.

CD45 isoforms differ by alternative splicing, affecting extracellular domains but sharing identical transmembrane and cytoplasmic regions. Expression varies by cell type and physiological state. CD45 also promotes cell survival via integrin-mediated signaling and participates in apoptosis-related DNA fragmentation. It is expressed on all human hematopoietic lineage cells except mature red cells. Dysfunctions are associated with severe combined immunodeficiency and Hepatitis C.
