Product Specific Information

The SJ25C1 monoclonal antibody reacts with human CD19, a 95 kDa transmembrane glycoprotein expressed on B cells throughout development except for terminally differentiated plasma cells. It is also expressed on follicular dendritic cells. CD19 forms a multimolecular complex with CD21, CD81, Leu13, and MHC class II, associating with the BCR to mediate signaling that induces tyrosine phosphorylation, calcium flux, and B cell proliferation. SJ25C1 and HIB19 recognize overlapping epitopes.

Applications

• Reported for use in flow cytometric analysis
• Tested by flow cytometry using normal human peripheral blood cells
• Recommended usage: 5 µL (0.8 µg) per test in 100 µL final volume
• Suitable for 10⁵–10⁸ cells/test
• Antibody titration recommended for optimal performance

Important Notes

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI)
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining
  • Add 5 µL per sample before combining with cells
  • For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells)

Target Information

CD19 is a member of the immunoglobulin superfamily with two Ig-like domains. It is expressed on 100% of peripheral B cells and on certain myeloid leukemia cells. CD19 functions as a signal-amplifying co-receptor for the B cell receptor (BCR), regulating B cell activation and proliferation. Mutations can cause hypogammaglobulinemia, while overexpression leads to B cell hyperactivity.
