Target Information
STAT4 was originally identified using degenerate primers complementary to conserved regions of other STAT proteins. It is most similar to STAT1 (52%) and STAT3 (47%). Functionally, STAT4 is phosphorylated by Jak1 or Jak2, forming homo- and heterodimers with other STATs. Phosphorylated STAT4 binds the IFN-gamma activated site (GAS), and serine phosphorylation is needed for maximal transcriptional activity.
STAT4 expression is restricted to thymus, spleen, and testis. It is activated by interleukin 12 (IL-12), which induces IFN-gamma production and promotes Th1 cell development. STAT4-deficient mice show impaired Th1 differentiation and enhanced Th2 development. STAT4 activation by IFN-α/β is also required for IFN-γ production during viral infection.
