Target Information
The MAP kinase-interacting kinases (MNK1 and MNK2) are activated by MAP kinases and regulate protein synthesis by phosphorylating eIF4E on Ser209, potentially contributing to tumorigenesis. Overexpression of Mnk2 increases phosphorylation of endogenous eIF-4E, indicating its role as an eIF-4E kinase in vivo. Mnk2 may be involved in responses to environmental stress and cytokines. Expression of active MNK1 and MNK2 mutants in 293 cells reduces cap-dependent translation compared to cap-independent translation. Human Mnk2 shares approximately 94% identity with murine Mnk2 and 71% with human Mnk1. In vitro phosphorylation studies show Mnk2 is a better substrate than Mnk1 for Erk2, p38MAPKα, and p38MAPKβ.
