Target Information
Ki-67 is a nuclear protein expressed during various stages of the cell cycle, especially in late G1, S, G2, and M phases. It associates with euchromatin at the perichromosomal layer, centromeric heterochromatin, and nucleolus via its forkhead-associated (FHA) domain. The protein shows cell cycle–dependent localization: perinucleolar in G1, nuclear matrix in G2, and chromosomal during M phase. Ki-67 is widely used as a proliferation marker since it is absent in G0 cells and increases from G1 through mitosis. Ki-67 antibodies help determine the growth fraction in neoplasms, with prognostic value comparable to thymidine-labeling index. Low Ki-67 index correlates strongly with low-grade tumors, and it is routinely used as a neuronal marker of cell cycling and proliferation.
