• Immunohistochemistry (PFA fixed):
  Incubate antibody for 30 minutes at room temperature.
  Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

8-hydroxyguanine은 활성산소에 의해 유도된 산화적 DNA 손상 형태로, G:C → T:A 전환을 유발합니다.
E. coli에는 이러한 돌연변이 효과를 방지하기 위한 세 가지 DNA 복구 효소가 존재하며, 그중 MutM은 효모(yOgg1) 및 인간(hOgg1) 상동체를 가집니다.
hOgg1 단백질은 DNA 내 피리미딘 반대편의 8-hydroxyguanine을 효율적으로 제거하고, 세균 및 효모 효소와 유사한 방식으로 AP 부위를 절단합니다.
손상된 DNA 복구를 조절하는 유전적 배경은 암 발생 감수성과 관련이 있으며, hOgg1 유전자는 다양한 암에서 이형접합성 소실(LOH)이 관찰되는 3p26.2 영역에 위치합니다. 특히 3p25–p26은 폐암에서 공통적인 LOH 영역으로 알려져 있습니다.
