Target Information
FGF2 (fibroblast growth factor basic, FGFb) belongs to the fibroblast growth factor (FGF) family and interacts with high-affinity transmembrane receptors to regulate cell proliferation and tissue neovascularization.
FGF2 exists as five isoforms with distinct intracellular localizations and functions:
- 18 kDa isoform: predominantly cytosolic, acts through cell surface receptors
- 22, 22.5, 24, and 34 kDa isoforms: nuclear, may signal independent of transmembrane receptor pathways
In humans, the FGF2 gene is located on chromosome 4q. It is involved in limb and nervous system development, wound healing, and tumor growth.
FGF2 mRNA contains multiple polyadenylation sites and is alternatively translated from non-AUG and AUG codons, producing five isoforms with distinct properties:
- CUG-initiated isoforms: nuclear, responsible for intracrine signaling
- AUG-initiated isoform: cytosolic, responsible for paracrine and autocrine signaling
Diseases associated with FGF2 dysfunction include Kaposi Sarcoma and corneal neovascularization.
