Product Specific Information

The monoclonal antibody 2TL242 recognizes human TAL-1. The TAL-1 protein is encoded by the tal-1 (T-acute leukemia or stem cell leukemia (SCL)) gene and functions as a 42 kDa basic helix-loop-helix transcription factor involved in mediating cell growth and differentiation.
Rearrangements of the tal-1 gene are common in human T-cell acute leukemias (T-ALLs) and result in aberrant transcription in leukemic cells. TAL-1 expression in leukemic cells is localized to the nucleus, although both nuclear and cytoplasmic expression has been observed in normal cells such as erythroid precursors, megakaryocytes, endothelial cells, and macrophages within tissues like tonsil, cerebellum, kidney, lung, and spleen.
TAL-1 regulates embryonic and adult hematopoiesis and has been found to influence proliferation and differentiation of common myeloid precursors to monocytes and macrophages.

Applications Reported:
This 2TL242 antibody has been reported for use in western blotting, microscopy, immunohistochemical staining, and immunocytochemistry.

Applications Tested:

  • Tested by immunohistochemistry of formalin-fixed paraffin-embedded human tissue using low or high pH antigen retrieval at ≤20 µg/mL.
  • Tested by western blot of reduced Jurkat cell lysate at ≤5 µg/mL.
    It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.

Target Information

TAL-1 is a basic helix-loop-helix transcription factor that regulates differentiation and survival during hematopoiesis. It is implicated in the genesis of hematopoietic malignancies and plays a role in erythroid differentiation. Mutations are associated with T-cell leukemia and melanoma. TAL-1 binds to the LIM domain-containing protein Rhombotin-2.
