Product Specific Information

The monoclonal antibody eBio8D9 reacts with an epitope shared by mouse CD21 (CR2) and CD35 (CR1). These are alternatively spliced transcripts from the Cr2 gene, producing surface proteins of 145 and 190 kDa, respectively.
CD21/CD35 are expressed by mature B cells, but not on thymocytes, peripheral T cells, erythrocytes, or platelets. Some evidence suggests expression on macrophages.

CD21 acts as a receptor for complement component C3d and Epstein-Barr virus (EBV). In association with CD19 and CD81, CD21 participates in B-cell activation. Cr2-deficient mice show impaired inflammatory and humoral immune responses in vivo.

The clones eBio4E3 and eBio8D9 do not cross-block each other, indicating they bind different epitopes.

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes at ≤ 0.125 µg/test (defined as staining a 100 µL cell sample). Optimal titration recommended.

Light Sensitivity: Tandem dye sensitive to photo-induced oxidation; protect from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-Step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark.
Excitation/Emission: 488–561 nm / 775 nm; compatible lasers – Blue, Green, Yellow-Green.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

CD21 (Complement Receptor 2, CR2) binds C3 complement fragments and plays a role in uptake and retention of immune complexes, memory B cell survival, and humoral response development. It also serves as a receptor for EBV and prion targeting.
A soluble form (sCD21) retains ligand-binding ability.

CD35 encodes a membrane glycoprotein found on peripheral blood cells, glomerular podocytes, and follicular dendritic cells. It binds complement components C3b and C4b and regulates complement cascade activity.
Variation in CD35 forms the Knops blood group system, with alleles F and S differing by 8 exons. A secreted plasma form has been described.
