Product Specific Information

The DX2 monoclonal antibody reacts with human CD95 (Fas, Apo-1), a 40–50 kDa member of the TNFR superfamily. CD95 is expressed by a broad range of hematopoietic and non-hematopoietic cells, including monocytes, neutrophils, lymphocytes, and fibroblasts. Interaction of CD95 with FasL induces apoptosis and plays a role in peripheral tolerance. DX2 does not block EOS9.1 binding, another antibody specific for human CD95.

DX2 monoclonal antibody is also reported to recognize canine CD95.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometry on normal human peripheral blood cells. Use at 5 µL (0.5 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).
Excitation: 488 nm; Emission: 520 nm; Laser: Blue Laser.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

FAS (Fas, CD95, APO-1) is a 46 kDa transmembrane glycoprotein functioning as a cell death receptor in the TNFR superfamily. It also exists in soluble form (~26 kDa). FAS stimulation aggregates FADD domains forming the DISC complex, activating caspases and triggering apoptosis.
FAS can activate NF-κB, MAPK3/ERK1, MAPK8/JNK, and participates in proliferative signaling in fibroblasts and T cells. Multiple splice variants exist, with soluble isoforms potentially regulating apoptosis negatively.
The Fas/FasL system is implicated in diseases such as AIDS, hepatitis, and cancer, and plays a crucial role in antiviral immune responses.
