Reconstitution:

• Reconstitute leptin in sterile, distilled water to a concentration of 1.0–5.0 mg/mL.
• Further dilutions should be made in aqueous buffers supplemented with carrier protein (e.g., 0.1–1.0% BSA).
• Briefly centrifuge vial before opening to collect contents at the bottom.

Storage:

• Lyophilized preparation is stable at room temperature for up to 3 weeks.
• For long-term storage, keep at -20°C, preferably desiccated.
• After reconstitution, stable for up to 1 week at 2–8°C.
• For maximal stability, aliquot and store at -20°C.
• Avoid repeated freeze/thaw cycles.

Target Information

Leptin is a 167 amino acid protein hormone that regulates body weight, metabolism, and reproductive function. It is approximately 16 kDa and encoded by the obese (ob) gene. Leptin is mainly secreted by adipocytes, reflecting total body fat mass, and also produced in the stomach epithelium and placenta. Its receptors are highly expressed in the hypothalamus, T lymphocytes, and vascular endothelial cells.

Leptin influences feeding behavior, energy expenditure, thermogenesis, hematopoiesis, angiogenesis, wound healing, inflammation, immune responses, pubertal development, and fetal growth. It plays roles in obesity, anorexia nervosa, insulin resistance, and hypertension. Defects in leptin production can cause severe hereditary obesity in rodents and humans.
