Product Specific Information
- For immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
- For formalin-fixed tissues: Heat tissue sections in 10 mM Tris Buffer with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, then cool at room temperature for 20 minutes.
Target Information
TNF alpha is a multifunctional proinflammatory cytokine belonging to the tumor necrosis factor (TNF) superfamily. It is mainly secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF alpha regulates immune cell activity, proliferation, differentiation, apoptosis, lipid metabolism, and coagulation.
It exists as multimers (dimers, trimers, or pentamers) but appears as a single 17 kDa band under non-reducing SDS-PAGE conditions.
Knockout studies in mice indicate neuroprotective functions, and TNF alpha induces tumor necrosis in tumor-bearing mice.
It plays critical roles in immune responses to bacterial, viral, parasitic, and fungal infections, as well as in tumor necrosis.
TNF alpha is implicated in autoimmune diseases, insulin resistance, and cancer.
