Product Specific Information
The 90 monoclonal antibody reacts with the mouse CD38 molecule (~42 kDa type II transmembrane protein).
CD38 expression increases during B-cell differentiation and is down-regulated in germinal center B cells and mature plasma cells. It is also expressed on subsets of thymocytes, mature T cells, and NK cells. Crosslinking of CD38 induces B-cell proliferation, enhanced by IL-4 and LPS co-signals. CD38 acts as a counter-receptor for CD31 and functions as an ectoenzyme with cyclase and hydrolase activities, playing roles in lymphocyte activation and differentiation.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes at ≤0.5 µg/test (100 µL final volume). Optimal titration recommended.
Fluorochrome Information:
PE-eFluor 610 excitation: 488–561 nm, emission: 607 nm. Recommended filter: 610/20 band pass (PE-Texas Red equivalent). Ensure instrument compatibility.
Light Sensitivity: Protect vial and samples from light.
Fixation: Samples can be fixed using IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) and stored up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.
Laser Compatibility: Blue, Green, Yellow-Green lasers.
Target Information
CD38 (NAD+ glycohydrolase) is a type II transmembrane glycoprotein involved in activation, proliferation, and differentiation of lymphocytes, as well as apoptosis of progenitor cells. It functions as a multi-catalytic ectoenzyme (ADP-ribosyl cyclase, cyclic ADP-ribose hydrolase, NAD+ glycohydrolase) and as a cell surface receptor.
Antibodies to CD38 are used for:
- Subtyping of lymphomas and leukemias
- Detection of plasma cells (e.g., myeloma identification)
- Marker for activated B and T cells
CD38 also participates in cell adhesion, signal transduction, and calcium signaling. Expressed at high levels in pancreas, liver, kidney, malignant lymphoma, and neuroblastoma.
Associated diseases include chronic lymphocytic leukemia and Richter’s Syndrome.
