Product Specific Information
The M5/114.15.2 monoclonal antibody reacts with mouse MHC class II molecules (I-A and I-E subregion-encoded glycoproteins). It detects polymorphic determinants on B cells, monocytes, macrophages, dendritic cells, and activated T lymphocytes from specific H-2 haplotypes (b, d, q, p, r, u).
This antibody has been tested for flow cytometric analysis of mouse splenocytes, recommended at ≤0.4 µg/test (100 µL volume, 10⁵–10⁸ cells/test). Optimal titration is advised.
Each product includes:
- 1 vial NovaFluor conjugate
- 1 vial CellBlox Plus Blocking Buffer
Important Notes:
- NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability detection.
- CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
- Add 5 µL per sample before mixing with cells.
- For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).
NovaFluor conjugates employ Phiton™ technology, offering engineered fluorescent signatures with minimized spillover and spread.

Target Information
MHC class II molecules are transmembrane glycoproteins expressed on antigen-presenting cells (macrophages, dendritic cells, B cells). They bind endocytosed exogenous antigens and present them to T cells. The antigen-binding groove is open at both ends and accommodates peptides of 15–24 amino acid residues.
HLA and MHC antibodies are crucial in Immunopeptidomics for neoantigen identification via LC-MS/MS.
